    The antiserum does not cross react with any other component of human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. of this antiserum has not been tested in detail.

    Purification

    Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other plasma proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.

    Immunogen

    Alpha-1 lipoproteins are completely different from beta-lipoproteins. In fresh serum only one alpha-1 lipoprotein exists immunologically, but after centrifugation, storage, freezing and thawing two types of alpha-1 lipoprotein with difference in mobility and antigenic properties have been observed. Alpha-1 lipoprotein in fresh serum is usually precipitated by the specific antiserum as a rather symmetric arc in the albumin zone. Concentration in normal human serum is 170-320 mg/100 ml. Freund’s complete adjuvant is used in the first step of the immunization procedure.

    In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of alpha-1lipoprotein in human plasma or other body fluids or to determine its concentration. The presence of non precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive non-precipitating antibody-binding techniques if proper controls are included.

    Background

    The reactivity of the antiserum is restricted to alpha-1 lipoprotein. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified alpha-1lipoprotein.
